Introduction:
To produce beneficial effects in the gastrointestinal tract (GIT), probiotic microorganisms must have the ability to survive and metabolize in the intestine. Therefore, they must be resistant to the acid levels of the GIT.
Probiotic formulas must also contain a large number of viable organisms (highly concentrated) that, when ingested, survive the rigorous attacks of the gut. upper part of mammals for its bacterial content to reach the small intestine. One of the main barriers to the passage of bacteria is the acidity of the stomach. The pH of the stomach varies throughout the day under the influence of the buffering action that food or liquid may have on the stomach. However, the fasting pH of the human intestine is around pH 3.0.
The time it takes for food to pass through the stomach also varies widely, from a few minutes to an hour or more. The food itself will have some neutralizing effect on the pH of the stomach, and therefore a pH of 3.0 is
probably the lowest the bacteria will be subjected to. In addition, food can play a physical protective role.
Thus, the conditions of our laboratory tests will probably be the most vigorous conditions that the bacteria will be subjected to, i.e. a pH of 2, 0 for 2 hours, and the results obtained should be analyzed accordingly.
It should also be noted that the laboratory conditions (in vitro tests) are designed for these tests, although they try to mimic the
conditions inside the stomach, they are, in fact, a very complex situation.
When kept at pH 2.0 for a period of 2 hours, there was no significant loss in viability/concentration of any of the bacterial strains. Contact time is extreme: a two-hour period of contact without any buffering effect, for example, from food or water.
Conclusions: Total viable bacterial counts did not decrease in viability/concentration after contact with pH 2.0 acid for 2 hours. This means that a high concentration of microorganisms survive, reaching the small intestine and establishing themselves as part of the normal microflora.
